He went to the St. Thomas More School in Connecticut and later graduated from Calvert Hall College High School in Towson. In his final season, Lee got the First-Team All-Baltimore Catholic League selection and Second-Team Baltimore Sun All-Metro.

He started his basketball career with the Drexel Dragons, where he was the point guard in the first season and maintained an average of 12 points and 4.4 rebounds. His impressive start to his college career won the Colonial Athletic Association Rookie of the Year honors. However, he experienced an injury in a game against Arizona, leading to his exit from the season.

The next season, Damion Lee was given the Second-Team All-CAA acknowledgment after finishing third in the CAA with an average of 17.1 points per game. He stood fourth in three-pointers per game (2.3) and second in the CAA in free throw percentage (82.9%).

Damion Lee fractured his right hand at Northeastern and finished the game with 30 points. His injury caused him to miss the season’s last three games. Later in March 2015, he announced his transfer from Drexel University to Louisville for his final college season in 2015 – 2016. Lee became the top point scorer for Louisville in 2015. He was also listed on the 35-man midseason watch list for the Naismith Trophy in February 2016.

Lee went undrafted in the 2016 NBA draft and joined the Miami Heat for the 2016 NBA Summer League. He moved to Boston Celtics in September 2016 and appeared in two preseason games for them. While the Bolton Celtics waived him, Maine Red Claws acquired him in October as an affiliate player of the Celtics. Damion Lee played 16 games for the Maine Red Claws and was traded to the Santa Cruz Warriors on August 24, 2017.

As his career grew, Lee signed a two-way contract with the Golden State Warriors on July 14, 2018. It led to his return to the Santa Cruz Warriors. He enjoyed a career-high 23 points and recorded a double-double in a 134–123 victory over the New Orleans Pelicans. Lee bagged a multi-year contract with the Golden State Warriors in January 2020. He scored 12 points with a three-pointer leading to a 129–128 win over the Chicago Bulls on December 27, 2020. He won the 2022 NBA Finals with the Warriors.

In July 2022, Damion Lee joined the Phoenix Suns and put in 11 points, two assists, and two rebounds in his debut match with the Suns.

Besides NBA, Lee also had the honor to become a part of the US national team in the 2019 FIBA Basketball World Cup qualification, replacing Marcus Thornton.
